Mode of Action:
The mode of action of Quetiapine is unknown. However, it has been proposed that this drug's efficacy in schizophrenia is mediated through a combination of dopamine D2 receptor antagonism in the mesolimbic pathway and serotonin 5HT2A receptor antagonism frontal cortex. In bipolar depression and major depression, Quetiapine's actions may be attributed to the binding of this drug or its metabolite to the norepinephrine transporter. Quetiapine's agonism of serotonin 5HT1A may relieve anxiety & depression, antagonism of histamine H1 receptors may explain the somnolence and that of adrenergic ?1 receptors may explain the orthostatic hypotension observed with this drug.
