Itraconazole
Description:
SUBA (Super Bioavailability) technology is a novel technology for enhancing the bioavailability of poorly soluble drugs. The technology utilizes a solid dispersion of drug in a polymer that improves the dissolution of poorly soluble drugs compared to their normal crystalline form. Itraconazole is an orally active triazole antifungal drug that has demonstrated a broad spectrum of activity and favorable pharmacokinetic profile. Itraconazole inhibits cytochrome P450 dependent enzymes resulting in impairment of the biosynthesis of Ergosterol, a major component of the cell membrane of yeast and fungal cells.

Indications:
Tinea corporis, Tinea cruris, Tinea pedis, Tinea manuum, Vulvovaginal candidiasis, Fungal keratitis, Pityriasis versicolor, Oropharyngeal candidiasis, Onychomycosis, Histoplasmosis, Systemic Infections: Aspergillosis, Candidiasis and Cryptococcal infection when other drugs are ineffective. Antifungal prophylaxis: To prevent systemic fungal infection in severely neutropenic patients.

Mode of Action:
The mechanism of action of itraconazole is the same as the other azole antifungals: it inhibits the fungalmediated synthesis of ergosterol, via inhibition of lanosterol 14?-demethylase. Because of its ability to inhibit cytochrome P450 3A4 CC-3, caution should be used when considering interactions with other medications
